Abstract (English):
The new expression system based on the strain B.megaterium PV370 was constructed. The gene of new RNAse from Bacillus sp. (RBT) was cloned under the control of strong Pspac promoter, transcription of the gene was induced by isopropyl β-D-1-thiogalactopyranoside (IPTG). The new plasmid system allows us to reduce the time of synthesis of the enzyme and to increase the yield of enzyme production by several times in comparison with the natural producer. The results of this work may provide a basis for the subsequent isolation and purification of the recombinant protein RBT in preparative quantities to study its biological properties.
